ASK1 antibody - (Feb/07/2007 )
Hi,
I just would like to know if anybody has ever used ASK-1 (apoptosis signal-regulating kinase-1) antibody. Because I need to use this antibody for some selenium induced cells to see this protein activation promoting apoptosis and when I search for papers, everybody use different forms of it (phoshporylated at different sites),and I'm confused! I don't know which antibody is exact for my research?(which phosphorylated site) Please help me with this problem.

It depends in which signalling pathways you are intereste. I here quote from a Cell signalling data sheet, I am very satidfied with their ASK1-Ab:
"Phosphorylation at Ser967 is essential for ASK1 association with 14-3-3 protein and suppression of cell death (6). Oxidative stress induces dephosphorylation of Ser967 and phosphorylation of Thr845 in the activation loop of ASK1, and both are correlated with ASK1 activity and ASK1-dependent apoptosis (7,8). On the other hand, Akt phosphorylates ASK1 at Ser83, which attenuates ASK1 activity and promotes cell survival (9). Phosphorylation at Ser967 of ASK1 leads to association of 14-3-3 and suppression of ASK1 kinase activity (6)."

Thank you very much The Bearer. I used the one phosphorylation of Thr845 and it works great!!!!
