Codon Usage in Yeast 2 Hybrid - (Feb/03/2007 )

I am using clontech yeast 2 hybrid system and have cloned a human gene into the vector. However, there are 10 codons in which 40% of Leucine are changed to Serine. Is it a serious problem?

If yes, will the problem be solved when I use the yeast strain AH109 from clontech, is there already tRNA replacement in this strain?

about your first question generally leucine is hydrophobic amino acid while serine is hydrophobic (non-polar) and hydrophilic (something between) but polar and uncharged this means it really depends where this amino acids are within your protein. there are some hot point that even a simple change couse serius problem a change in Iso-leucine to valin in Hemoglobin causes sickle shape Hemoglobin and sever anemia. but in some cases change in protein even 30% of total amino acids causes really nothing.
