melanin extraction - (May/05/2003 )

Probably more of a molecular question....however, does anyone know of a protocol for melanin extraction (from a melanoma cell line like B16/F0) - that has been successful and replicable??

Hi, Im culturing B16F10 cell line. I need to to extract the melanin from the cells as well. I have tried by using 1N NaOH, incubate at 80 degree celcius for 1 hr. The melanin content release as the cells lyse. However, the color produced by the melanin content very light and cannot be measured by the microplate reader....it gives very low absorbance value at 400nm (OD = 0.25-0.35). Does it mean I need to use more cells?

Anybody knows abt the method to determine melanin content from B16F10 cells?
