Hi all
will it be sufficient enough to use 32 or 34 degree annealing temperature for RAPD PCR using 10 mer primer from Operon? or do we have to adjust the temperature according to the sequence?

For RAPD you do not care about the sequence, you just adjust the temperature depending on how precice you want the priming.
I have never tried annealing temp below 38, and have most often found 40-42 C beeing better for gaining reproducible results.
I can however not rule out the possibility of some compound put into the reaction, that will make it possible to get specific primig at even lower temperatures.
