how to count the rounded cells in the well? - (Jan/29/2007 )

Hi everybody,

I'm trying to do Y1 adrenal cell assay and should count the rounded cells after adding cholera toxin.
Please anyone tell me how to count the rounded cells in the well?

wouldn't you just put them under the scope and count the # rounded/field at a given magnification? perhaps also count the unrounded, and express as a percentage?

I've tried to count the rounded cells under microscopy but it seems difficult because the Y1 cells tend to clump together ... just wonder if there is an alternative method ... Do you think I can recover the cells by flushing with PBS?

possibly, but I think your error would be high.

I'm trying to check binding capacity of my protein to GM1 ganglioside receptor. Please tell me if you have any idea.
