Hygromycin B concentrations to select infected Blin-1 cells - Which concentrations of Hygromycin I should use to select infected Bli (Jan/29/2007 )
Hi everyone,
I infected Blin-1 cells with retroviruses carrying a Hygromycin resistant gene. I used Hygro B to select them. But the control cells still survived at 400 ug/ml Hygromycin B. Please give me your expert advice about which concentrations of Hygro B I should use to select infected Blin-1 cells.

what control means? mock transfected? or untransfected? firstof all you need to know the Hygr B concentration that kills your untransfected cells. if is 400 ug/ml is a huge amount of antibiotic, I use to add between 50-250 ug/ml, depending on the cell type. have you know ifyour cell line is Hygro B resistant?

My control cells are uninfected. Actually, the normal range of Hygro B is 100 - 800 ug/ml for most of cell lines. I have also asked other people, and they suggested me to do a titration. Thank you for your reply.
