gene expression study - between treated and untreated cells (Jan/22/2007 )

hi all.

need help here.

I am studying the gene expression for my cell line in treated and untreated condition.
I measured the gene expression level with real time pcr with cDNA made.

Then I substract the Ct value of treated with untreated to get the difference.
Is this correct to get the difference in gene expression?

However, to normalize the gene expression, you should have a gene (s) such as GADPH, which are used as a control and will remain unchanged in response to the stimuli you are using.

As endogenous gene you can find also:
TBP, GAPDH, B2M, GUSB, HMBS, PGK1, PPIA, HPRT1...
I'm use to test all my endogenous gene on the cDNA treated's cells and untreated's cells to choose the best one. The best one will be on each plate of qRT-PCR as control.
