DNA conc for PCR - (Jan/18/2007 )

Please advise what the optimal concentration is for PCR amplifying genomic DNA. I have a stock conc of 0.3ug/ul DNA and generally do the reaction in 25ul. When I used 1ul of the stock, I got smears only. Is this too much DNA?

i use 100ng of genomic DNA, and it works well. could also the the temps of your reaction... or the primers.
