microtome - thin sectioning (Jan/14/2007 )
hi people,
I am new to microtome. I need to do some collagen staining. In general , it is said that for tissue staining the thickness allowed is 2-25 micron meter. We have a ultramicrotome in our lab (for electron microscopy), which cuts in nano thickness 100 nano. I am not sure whether if we cut it too thin, will it be still fixed to the microscopic slide and stained with different tissues. Pls help.

That will require plastic embedding and the removal of the plastic resin. Its much more time consuming. However, you can get very good resolution of the sections. You can cut thick sections. like 0.5 um to 2 um thickness.
