Human PBMC Vs Porcine PBMC - separation - (Dec/21/2006 )

I can separate Human PBMC from Human Blood without any trouble but trying to separate Porcine PBMC from Porcine Blood was not that successful.

What are the changes we have to do to the protocol? . . At what speed do U centrifuge Porcine Blood and for what time? Had to centrifuge at 3000rpm for 10 mins for the blood cells to clear from plasma. unsure.gif I wonder if that affected the cell viability.

We normally do 200 g but for longer time (20-30 minutes) BTW rpm does not tell us much, if we dont know the size of the rotor. So better to convert it to g (RCF).
