Is it possible to run a gel using plasmid DNA that already has primers in it and is restriction digested? Or is this usually done PCR instead of RE digestion? Thanks.

I'm not sure what your goal is...I run my restriction digest mixture on a gel to gel extract all the time, but if I'm not doing a digest, I will PCR purify before running my gel. Does that help?

well it's possible to run a digested PCR product with primers, if one of the expected bands are not around 100.
If one is 200, a 3% agarose gel can efficiently discriminate between it and primers.
