cell culture - (Dec/08/2006 )

When I culture cells to do RNA extraction at 48 hrs, is it OK to add fresh media in between (say, around 24 hrs.?) Or, should I just leave it like that? When I did the experiment, I added media at 24 hrs . but there was difference of opinion between 2 staff -it is ok vs. not ok. Any thoughts?

I think it is not necessary to change the medium.

I agree. If medium seems ok it's not necessary change it.

You are saying you extract RNA at 48 hrs; I assume that means your are adding some compound or something of which you want to see effect at the RNA level and after 48hrs of that (compound?) addition, you extract RNA. If this is the case I would leave cells in presence of a compound without changing the medium. You might want to add more volume of medium to prevent cells from starving.

what do u mean by u add media at 24 hours??/
i first grow fungal mycelia in a culture mediam then after 48 hours i transfer growth/mycelia in induction medium........is it the same????
