How to isolate intact mitochondria from live cells? - (Dec/06/2006 )

Hi.
I would like to ask, if anybody knows some reasonable protocol for isolation of mitochondria from a cell line in such a state that it will be possible to study an effect of a drug on mitochondria preparation. I am thinkink about measuring membrane potentials (but maybe there are some other methods).

Another question if it is possible to measure such phenomenona in 96 wells (so probable massive ammount of cells is needed).

Hmm. One more think - I found a message dealing with mitochodria isolation in general. So is there a way how to isolate cancer cells mitochondria without ultracentrifuge? Tx.
