mobility of protein band over PAGE - please suggest (Nov/23/2006 )

hi all,

i am resolving the HepG2 protein over the 12%resolving. but protein are not moving to the

whole length of the gel. as my gel is 5.5 inch and my band comes only within 2.5 inch.


Does any one have solution for this problem.

there are different reasons possible; two prominent possibiöities are:
there could be a leakage of current or cathode buffer, respectively; check if there is appropriate electrolytic gas bubbling during run;

another prominent reason is too much salt in probes or stacking/separation gel buffer
