Measuring CO2 levels in incubator - (Nov/22/2006 )
Hi,
Our lab recently shifted all cultures to another incubator (used one). As soon as we shifted, the cells stopped growing. I use J774 macrophages and the cells attach but they dont proliferate. We think the %age of CO2 in the incubator is the culprit. Is there any way I could measure the CO2 level in the incubator with enough accuracy ?. We are thinking of using Fyrite Gas analysers, but wanted to know whether its accurate enough or if someone has used it before.
Thanks,
Rao

Yea, they should give you a fairly accurate reading. We use a fyrite gas analyser in our lab and it does the job.

Dear Prao,
The Fyrite is accurate to +/- 0.5% and is the recommended way of measuring CO2 in incubators. J774 grow like weeds and they proliferate in just 5% FCS. The lack of growth could be due to :-
Contamination
Lack of CO2
FCS quality
Temperature
