Phosphate Buffer to adjust pH in LB - (Nov/20/2006 )
Has anyone used phosphate buffer to pH LB? If so, in what amount?

We just use 1 N NaOH (sometimes we dont even do that, E coli grow just fine). Unless you want to make Terrific broth (TB), no one bother to use phosphate buffer.

that is true, there is no change in pH with LB cultures adjucted with NaOH. The LB media is too poor for the E. coli culture to throw off enough acetate to change the pH significantly.

Hi, zera. I think you don't need to add phosphate buffer into your LB. Non-fastitious organisms like E. coli will still grow well in pure LB.
