plasmid purification from mammalian cells? - (Nov/16/2006 )

this may be a silly question!!!!!!!

is it possible??


is there a method out there whereby i could purify a plasmid from transiently transfected HEK cells??

would it be similar to a miniprep??

basically, as i know,
...a plasmid can't replicate in a cell.
...the cell degrade foreign DNA
...if you want conservation of the DNA, you need to apply antibiotic pressure, and thus the plasmid is opened (if not previously oppened ) and integrated in the genome

If you can develop a universal method for this,
that's a definitely a nature paper,

I think it's possible. Purify total DNA and transform into e. coli. See if anything grow.
