Cytotoxicity assay in THP-1 - In the refernce to infection (Nov/16/2006 )

Hi,
iam PhD student working on the drug delivery in the case of mycobacterium infection.
I want some clue to cytotoxicity assay after in vitro infection in THP-1 cells by crystal violet assay and LDH assay too.

Hi,
First of all thanks a lot, In our experiment its showinng increasing cells growth i mean no cytotoxicity after infection with mycobacterium its may be due to some problems in my experiment. I follow following protocole for this experiment:
Take THP-1 cells differentiated them by using PMA.
after 24h gives infection for 2h at 20MOI.
after 2h removed extracellular bacteria by removing media and add fresh media.
after different time point acces cell cytotoxicity by using crystal violet dye.
now fix cells by methanol for 1min and remove its
after washing with 1xPBS added .75% dye for 3min.
after incubation time completed wash 2-3 times with 1xPBS
dissolved left dye by 1% SDS and take reading by using ELISA plate reader at 595nm.
But in infected group showing maximmum viabil cells as comparision to normal one.
