Cell Culture with Human Plasma - (Nov/09/2006 )

The addition of human plasma to my cell cultures seems to be causing cytotoxicity. I use heat inactivated fetal bovine serum at a concentration of 10% and supplement with the human plasma on top of this (upto a total of 20% total serum). Any suggestions as to why the cells are so unhappy in human serum and how i could avoid this would be greatly appreciated.

Should'nt plasma be heat inactivated too? it contain also complement system.

well i thought only one of the two needs to be inactivated and i chose the fetal bovine serum as i am hoping to look for the toxicity of plasma from patients with Alzheimer's disease in the future (after i can get the controls to work!!) so heat inactivating the human plasma may cause some of the toxic proteins in such samples to lose their effect

but i will try heat inactivating both plasma to see the effects...thnx a lot
