help - stop codon (Oct/25/2002 )

hi,

I want to isert a fragment of my protein in pcDNT/tag. the problem is that at the end of this fragment there is stop codon. so my question is:
should I take of this stop codon when i design the reverse primer? because i 'm afraid that my protein will be cleaved from the tag.

Sure. You must take off this stop codon, or else your protein will be expressed without the tag.
