PFGE equipment - (Oct/20/2006 )
Morning all,
We're setting up BAC recombination in our lab at present and require a pulse field gel electrophoresis system to separate large fragments of DNA. I was just wondering if I could get any advice on what supplier/system to use? We only need standard conditions (i.e angle of 120 degrees) so the Biorad DR2 system has been suggested. I was just wondering if anyone on here had any good recommendations?

I myself used the Rotaphor system from Biometra quite sucessfully in the past. It's a rotating field PFGE machine. Not that I needed the "extra" angles it can provide, as I also did only BAC analyses, but the machine was already there in the lab, so I used it. Worked quite well for me....

Thanks mate, it's been between that and the biorad. Think i'll try and play the reps off against each other and see who offers me the best price!

We used CHEF from BioRad. It worked well and mapper was friendly:)
Note: Some people complain about the longevity of the electrodes.

true, they can be easily damaged
Hi, We also use the Chef DR-II from Biorad and have had no problems with the electrodes. Maybe we are lucky!
