Cell viability assays fortissues - (Jan/27/2001 )

For the time being we are trying to develop an in vitro system in which we can grow the tissues (instead of cells). But at this stage of our research, we need an assay to demonstrate that the individual cells of the grown tissue are still in a living state.Tryphan-blue exclusion assay, which is most widely used for asking the viability of the cells in suspensions, did not give good answers to us, since we are using tissue samples instead of suspension cells.And also the assays which determine the cell viability based the cellular proliferation is not useful our system, because our tissues contain also some types of non-proliferative cells.

If there is someone there, who have an idea for such an assay that we can use in our system, please share your idea with us.

Hi,Try the MTT-Cell viability test. It is often used with tissues and 3D cell cultures. MTT is a substane that is cleaved by mitochondrial Enzymes into an insoluble Formazan salt. This salt can be solubilized with acid Isopropanol. Perhaps you will find some protocols in the medline.
