How to make patch plate? - (Oct/14/2006 )
Hi, guys, can anyone tell me how to make the patch plates so i can do colony PCR while at the same time, keep the backup for later bacteria growth?

I would do it this way but I am sure there are other ways. Using a toothpick or tip (both sterile), pick your colony and patch onto a plate. Use the same tip to 'seed' a PCR tube. There will be enough remaining cells to lyse in the PCR tube in the initial denaturing step to release DNA for PCR.

Thanks, ML1975! But do you need to mark the plate since you are probably going to pick not only 1 colony for PCR?
