How to purify the ChIP DNA - (Oct/13/2006 )

hello everyone:

I am having trouble with the purification of ChiP DNA. The A260/A230 of ChIP DNA following by phenol/chloroform extraction is always only about 0.6. How can I purify it? I've tried using Qiagen PCR product purification kit. But by doing that, it waste too much ChIP DNA. Are there any suggestions?

I use DNeasy Tissue Kit from Qiagen and so far I had good recovery and no problems. I have to say I didn't use anything else therefore I cannot compare but from 1 million cells for each IP (with one Ab) I recover 200ul DNA and I use 2,5ul for each real-time reaction. I wouldn't change it wink.gif

Have you tried micro spin columns. They have a smaller capacity but you probably have a small amount of DNA in your ChIPs so that means you'll lose less.

Take a look at Qiagen Mini-elute. I'm sure there are others which may be cheaper.
