antibiotic selection for breast cancer MDA-MB-436 - (Oct/11/2006 )

Does anyone use G418 selection for post-transfected MDA-MB-436 cells? If so, what concentration should you use to get optimal selection? And, if you set up a kill curve, do you do that using normal, untransfected cells? Also, do you leave the cells about 48 hours post-transfection to get the G418 expressed before adding it to the media?

try a conc. curve using G418 for ur untransfected cells. u could try 0,200,400,600,800,1000 ug/ml.

imp. thing is u should know the active conc. of ur G418.

we start selection with antibiotics 48-72 hrs after transfection.
