look for protocol of glycero-gradient ultracentrifugation - (Oct/10/2006 )

Hi, everyone:
I want to use glycerol-gradient ultracentrifugation for fractionlation my cell lysate. but I don't know how to do it. Would anyone provide me some protocol or any experience?

It is easy to do. It is just time consuming.

Lysis buffer may be different upon different protein.

Just a little tip for you: U may make 50% glycerol by mixing which with your lysis buffer.Otherwise it will be very hard to get right volume of glycerol. U may make master mix for different layer and then add to your centifuge tube layer by layer slowly.
