Tobacco nuclei extraction from plant tissue - Nuclei extraction (Oct/05/2006 )

Hello all,

i'd like to know if anybody has a good protocol for extrating nuclei in tobacco. I have tried several but it seems that my nuclei were not pure enough, i got a lot of cell debris. Please, help!

try to adjust your centrifuge time this is very important stage.

Could you tell me what protocol are you following? at least, i can start from there!
