DNA Extraction from blood of Horseshoe Crab - Extracting DNA from the blue blood (Oct/05/2006 )

Hello all...

I'm currently facing difficulties in extracting DNA from the blue blood of horseshoe crab. I have tried many different protocols and even modifying some myself but none seems to be giving me any genomic DNA. What I got is just degraded DNA. I have already tried Sambrook's Phenol:Chloroform method and Aljanabi's salt precipitation method but at the moment none of these are working...

Any suggestion of methods or protocols from the members?

i would try calling qiagen, or zymo, or invitrogen, etc. and asking if any of their kits would work... then you could just model your protocol after that kit. i've had some trouble myself with the phenol chloroform. do you have the funding for a kit? those come with tech support, and a refund if it doesn't work. smile.gif
