How to isolate RNA from zebrafish tissue? - (Sep/30/2006 )

I have tried to isolate total RNA form several zebrafish tissues. Sinece zebrafish is very small, it took me a lot of time in tissue separation. I always can't' get a good ratio of 28S to 18S is 2:1.
Does anyone know how to improve it? (I usually use liquid nitorgen to sacrifice zebrafish) laugh.gif

I have used the RNAzol reagent many times and have had success. I used a glass homogenizer to lyse the tissues on ice and followed the standard protcol. I have used the RNA for microarrary and qRT-PCR successfully.
