My PCR product is 100bp (approx). What result is more reliable-gel or hybridisat - (Sep/27/2006 )

Hi,
What is the more reliable result to check for a 100bp PCR product.
A 6% gel or a hybridisation on membrane.

6% polyacrylamide?

What's wrong with a standard agarose gel? A 1% gel should do fine. Just run it at 100 V for 45 minutes (a little less than usual) and with a 50 bp ladder.

Or a 3.5% agarose, if one has to be extra careful and count the difference in 10s of bps.

Yeah, I guess it depends how clean you PCR is.

Hello,

If you know the sequence of the 100bp product than hyridization would clearly provide better specificity.
