Transformation efficiency of plasmid dna - (Sep/24/2006 )

hi,

how would the use of a plasmid mixture (different sizes) lower the transformation efficiency and frequency compared with a small, single plasmid only?

To the best of my knowledge, efficiency would drop mainly because large plasmids transform less efficiency then small plasmids. It takes a large plasmid more time to transit the pores formed during electroporation, while a smaller plasmid takes less time. Thus a small plasmid would transform more efficiently.

If there are other reasons, I am all ears.

this is what I was told as well. smaller plasmids better transformation.
