Long-time storage of tissues/cells with fluorescent tracers - (Sep/10/2006 )
What is to recommend for long-time storage of tissues or cells treated with fluorescent antibodies and mounted with Dabco-containing mowiol? At the moment we do not use additional preservations for the edge of the coverslips after polymerization of mowiol. Is it necessary (f.i. to protect against rehydration) and are there possibilities to seal the edge of cover slips?
So far, I haven't sealed the coverslips and my signal lasts for months using Dabco. I know some people use nail polish to seal the edges of the coverslips
thanks for advice; at which conditions do you store your probes? of course darkened but which temperature (RT, 4°, -20°, -80°C) you recommend?
4oC is fine
we kpet slides mounted with moviol in 4C. I have seen it preserve fluorescence for few months.
