Is it possible to do siRNA followed by transfection of dn of the protein? - HAs anybody tryed this?? (Sep/08/2006 )
I was wondering if its possible to knockdown a protein with siRNA and then transfect a dominant negative of that same protein?
Thank you

transfecting siRNA initially, they will degrade after a few days, so u could transfect ur protein.
I think there is a tet-on/tet off system to regulate a shRNA expression, this may help u for ur experiment.

That's what I thought, thank you very much!!

Since a DN protein usually is the truncated version of WT. You can select a siRNA against the part of the sequences unique in WY that does not exist in the DN mRNA, and co-transfect this siRNA and DN gene at the same time.
