pECL Eco viral vector and Rat cells - viral receptor present? (Sep/06/2006 )

Hi, I am trying to infect NRK ( rat) cells using a retroviral vector and pCL-Eco as packaging vector. I use 293T as producer line. My viral supernantant infects mouse cells (NIH-3T3) but is not working in NRK cells. According to the manufacturer of pCLEco (Imgenex), it should work in rat cells too. has anyone faced a similar problem. Can u suggest a reason and solution?

Try using more of the supernatant. May b it takes more time to express in these cells.

Could u change promoters in ur vectors? Sometimes having tissue specific promoter helps.
