ChIP control for transcriptional repressor? - (Sep/05/2006 )

Hi all!

I perform ChIPs with antibodies directed against a transcriptional repressor.

Antibodies against RNApol II don't seem very logical in this case, so does anyone of you has any suggestion, what positive control I can use?

Thanks a lot! biggrin.gif

Hello,

Again activemotif has provided the following infomation for negative ChIP control in their ChiP kit. http://activemotif.com/download/manual/chipit.pdf

QUOTE
ChIP negative control primers flank a region of genomic DNA between the GAPDH gene and the chromosome condensation-related SMC-associated protein (CNAP1) gene. The sequences are:
Neg For: 5´-ATGGTTGCCACTGGGGATCT-3´
Neg Rev: 5´-TGCCAAAGCCTAGGGGAAGA-3´
Product size: 174 bp

The negative control primers should give rise to equivalent amounts of product whether it is performed on DNA isolated by immunoprecipitation with anti-RNA pol II or with the Negative Control IgG
