You may find excelent protocols for T cell proliferation (both human and rodent) in Current Protocols in Immunology (Edited by: John E. Coligan, Ada M. Kruisbeek, David H. Margulies, Ethan M. Shevach, Warren Strober).
When searching for protocols this manual is a must for anyone doing immunology work.
