Protein lysis buffer - Choice of buffer for different proteins (Aug/31/2006 )

Hi! I'm now doing an immunoblotting for a membrane protein. And I have confirmed the protein exists in the sample by flow cytometry using the same primary antibody. But the result of western does not consist with flow cytometry result.

So, I think it maybe the problem of lysis buffer. Is there any online referrence of different protein lysis buffer for different use? Thanks

your problem may be that you denature your proteins for the western blot and the antibody is conformation specific.
