Selecting epitomes of antibodies to apply in Western Blotting - (Aug/29/2006 )
I need a little guide here...
I'm just wondering whether does anyone know which of these are more specific:
(1) epitome mapping near the C-terminus of a compound OR
(2) epitome corresponding to amino acids 65-164 mapping to an internal region of a compound
And why so??..

I guess you are trying to buy an antibody and you don't know which of the two you should choose... You should have a look at the datasheet of the Ab first to see whether it recognizes the epitope when the protein is folded or unfolded (that means if you can use the Ab for IP in native conditions or for WB to make it easy). If you want to study interactions, you should be careful not to choose for IP the Ab that recognizes a region of your protein that binds to its partners (or the partners you are interested in) otherwise you will IP only the fraction that doesn't bind the partners. You should choose which Ab is better for your application(s)
