multi shRNA to improve knockdown - (Aug/28/2006 )
Hi,
i was wondering if anyone has an opinion of using multi shRNA's in order to improve knockdown.
there's a paper (http://www.ncbi.nlm.nih.gov/entrez/query.fcgi?db=pubmed&cmd=Retrieve&dopt=AbstractPlus&list_uids=16869514&query_hl=24&itool=pubmed_docsum) which describes using multi miRNA hairpins to significantly improve knockdown. would this also work using shRNA?
is there a reason (perhaps due to dicer or drosha) that it would/wouldn't work?
i've only used one hairpin at a time, and had reasonable knockdown... but if this paper is true, why not use more?
just wondering what people thought about it.

it is possible for shRNAs. my friend succeded using lentiviruses with 2shRNAs. however, i've heard that sometimes you can observe some sort of "competition" between two shRNAs. i'm not sure how to explain this...
