restriction of Open Reading Frame (ORF) - (Aug/27/2006 )

Hi all....

Can anyone suggest something regarding this....
the open reading frame in the gene i have isolated from the genomic DNA lies 66-350bp...and the amplicon is 801bp. is it possible for me to restrict digest this ORF alone and clone into pichia pPICzalpha vector and transform Pichia pastoris for expression of the particular protein.....
do it fast....iam at stake....

If u have the respective sites on both the gene and in ur pPIC alpha vector, its possible. Else PCR amplify with the sites and then clone it.
