Nuclease free water for PCR - DEPC treated vs non DEPC treated (Aug/12/2006 )

hi guys!!

i juz want to know the difference btw these type of nuclease free water. which one give better results in PCR? and for sequencing also.. thanks unsure.gif

U use DEPC if you are dealing with RNA, I never heard of doing PCR with DEPC water, in some cases can even inhibit enzymatic activity

Normal a sterile water is used for PCR and even for preparation sequencing reactions One uses nuclease free when working with RNA, from cDNA onwards use the normal sterile water.

We use DEPC water for all things with RNA and everything else autoclaved or Millipore water.

DEPC is degraded by autoclave in alcohol substance and other stuff. Thus this alcohol may inhibit partially or fully the PCR reaction.

I use DEPC water for PCR and it words well

We use bidistilled water for PCR, no problems. It is not autoclaved and but frozen in aliquots. You can also buy nuclease free water from companies.
