Help Frozen lipofectamine - (Aug/09/2006 )

Hi i am really worried, i transported my lipofectamine from one lab to another in a dry ice container, and when i arrived to my lab and i open the box i found that the lipofectamine has been frozen i put it immediatly at 4 C, but i don't know if i damaged the lipofectamine!!!!!!!!! can i still use this lipofectamine, i need to do an Adeasy transfection tomorrow and i don't know if is going to work or not.




PLEASE HELP!!!!!

You can do a quick transfection with 293 using suspension transfection method as suggested by scolix, you should be able to see EGFP results by the time you do Adeasy tomorrow. You make suspension cell by trypsin/EDTA, wash with 10%FBS-DMEM, resuspend in serum free OPti-MEM medium, add cells to DNA/lipo 2000 mixture, change medium after 4 hrs, or do nothing. Check % green cells tomorrow, as late as you can wait.

No change to minor decline of transfection efficiency may occur. Tell us what do you find. Good luck.

thanks!!! i am going to do the transfectio with egfp taht i have and tomorrow before my Adeasy transfection i am going to check the fluorescence

Just recently I have a tube of lipofectamine 2000 frozen twice at -20C by mistake. I still used it in siRNA transfection and got what I expected.

I dont think freezing the lipofectamine should make such a big difference.

i had a very good transfection almost 90%!!!!!
