Thats because these are hepatocyte-derived. Acually, they are poor host cells for supporting HBV virus replication. Not sure what did you mean by DGD expression. Can you tell us the source of this work?

If I understood correctly, DCD is included in transfection as an internal control to qunatitate/normalize RNA expression level. Transfection is a technique known to vary a lot in different wells/dishes due to minute details of experimental procedure are done slightly differently. It is always good to have an internal control to correct the variation introduced solely by transfection. DCD is from duck and will not have endogenous mRNA similar to human RNA, therefore can be specifically probed by Northern blot. You certainly do not have to use the same DCD for your experiment, as the authors used it because they have it on hand.
