hybridima’s productivity - (Aug/07/2006 )

Hi all,
I have a human-human hybridoma, but unfortunately very lazy one.
It takes a lot of time to produce even small quantity of the mAb.
Is there any way to increase hybridima’s productivity?

Hi pery,
In which medium are you growing the clones in?
I think HT medium should be used (HAT without aminopterin) to maintain the secretion of the clones.
try it out.
It also could be because of mixture of secretors and non-secretors eventhough it is a monoclonal wherein the non-secretors overgrow the slightly slowgrowing secretors
I think you should be fast at this step.

Continued subcloning may or may not yield a better producer - there is always some variability in "clonal" population an some clones may make more mabs than others - I know in industry some lines are subcloned 4 or more times and hundreds of clones screened to isolate better producers- also supplementing with hybridoma cloning factor or extender ( I think brand name is Excite) may improve yield but it will cost you time and/or money.
