total protein assay in cell extracts - (Feb/16/2002 )
what is the best methods of total protein concentration in mamal cell extracts

i use the dc protein assay available from biorad. it is very quick, easy and reliable.

I have used the DC assay (based on the Lowry method) the Bradford assay (also available from BioRad or Pierce) and the BCA assay (Pierce). All in microplate format
My preference is the Bradford assay but the BCA works well also. I found the Lowry assay had the lowest sensitivity and I believe it has the largest protein to protein variation. One problem with the Bradford assay is that the filiter required for a multiwell plate reader (595nm) is not standard so you ma not have it already.

is there anyway to quantify or at least qualify the total protein concentration "in situ" ??
