dsRNA of EGFP as control? - (Jul/23/2006 )

Hi there,

I am currently working on a RNAi experiment in nematodes.
We use dsRNA of the target gene and dsRNA of maltose binding protein as a control.
Can I also use dsRNA of EGFP as a control?

You can: you can see it in some papers. But careful because I and other people from our lab had problems with a EGFPsiRNA (others from the forum didn't with the same one)

What kind of problems did you observe? I will use dsRNA of only 150 bp of EGFP. blink.gif

It decreased the expression of the cotransfected DNA. But other people in the forum didn't have this problem

May I ask what kind of problems you had?

I have EGFP siRNA, too, but I haven't tried it yet. Do you know how I can measure the efficiency of silencing? FACS is too expensive for us. We have a fluorimeter. Is it enough?
