alternative to dmso in pcr - (Jul/20/2006 )

hi! I'm looking for additives for my pcr because I can't use dmso as the ingredient. We want to analyse the product with dhplc (varian), that do not facilitate the use of any detergent and oil.

I don't think DMSO qualifies as a detergent or oil, so I'm a bit confused. Other additives are glycerol or betaine, in similar (2-8%) concentrations. You could also purify your DNA after the PCR by precipitation or PCR cleanup on a column.

if GC-rich "salt free" pure Tris-HCl buffer

Before trying to ADD something to PCR (betaine inosine etc) try to REMOVE something
for example KCl
