Is it possible: Effect of OVA on murine BMDC phenotyp ?! - (Jul/19/2006 )

Hi,

Does anyone knows, if OVA itself triggers a kind of differentation programme in DCs ?
I´m working with murine BMDCs. After culturing them for 6 days in 10mm dishes, the DCs are mostly floating around in suspension. But whenever I added 1mg/mL chicken Ovalbumin ( grade V from Sigma, reconstituted in destilled water ) to my DCs for overnight incubation, on the following day (when I add the LPS ) the DCs were already adherent !! Whereas those that didn´t see OVA where still unchanged.
In the FACS analysis there was no difference between the two groups after LPS stimulation. They were both MHC II high and CD86 medium.
Has somebody made similiar observations with OVA and DCs or could it be that the OVA is contaminated with some endotoxins ?

It is very likely that your OVA is contaminated with LPS. Especially Sigma OVA is known for high LPS contamination. OVA from Worthington is already way cleaner and would probably be fine. There is also some ultrapure OVA out there, but I can't remember the company.

in the meantime we have launched EndoGrade Ovalbumin with a endotoxin concentration of < 1 EU/mg (1 mg/ml). More information you find on www.endograde.de.
