how to culture human monocytes... - advice please (May/23/2002 )

i would like to culture some human monocytes to study glucose effects, does anyone have advice as to how i would succesfully culture them for approximately 4 days...?

I am also searching for the best growing conditions for the PBMCs. I am culturing them in RPMI 1640 medium for 24 hours. But still we are not satisified with this.

RPMI 1040 (plus L-glutamine and NaHCO3), supplemented with 10-20% FBS, 10% Human Serum, and antibiotics and antimycotics works well for up to 12 days. You should change media every 3 days...
