Heating during lysis - (Jul/17/2006 )

Dear Fellows
i want to ask you during protein extraction you keep the lysis step at room temperature or at some higher temperature.

when I lyse cells, I keep at 4°C to avoid protein degradation by proteases.

I'm with Missele. I always keep my cells (no pun intended) on ice. The detergent does the job for you. No need to heat the mixture up.

i always do lysis on ice/10 minutes.
if u can shake them on a rocker, it wud be efficient but not mandatory.
